aB-Crystallin in Lens Development and Muscle Integrity: A Gene Knockout Approach
نویسندگان
چکیده
METHODS. Gene targeting in embryonic stem cells was used to generate mouse lines in which the aB gene and its protein product were absent. Gene structure and expression were characterized by genomic Southern blot, immunoblot, and Northern blot analyses, and two-dimensional gel electrophoresis. The gene knockout mice were screened for cataract with slit lamp biomicroscopy, and dissected lenses were examined with dark-field microscopy. Lenses and other tissues were analyzed by standard histology and immunohistochemistry. Chaperone activity was determined by heating lens homogenate supernatants and measuring absorbance changes.

متن کاملDifferential Expression of aA- and aB-Crystallin During Murine Ocular Development
Results. During ocular development, aB-crystallin transcripts are present in the lens placode at E9.5. Transcripts of aA-crystallin are first observed in the lens cup at E10 to 10.5. During subsequent development of the lens, aA crystallin transcripts are most abundant in the fiber cells, and aB crystallin mRNA is preferentially expressed in epithelial cells. Transcripts of aAcrystallin were de...

The aB-crystallin protein, the predominant structural protein of the ocular lens, is a member of the small heat shock proteins that is also expressed abundantly in the heart and skeletal muscle.1 The aB-crystallin was initially discovered in the vertebrate ocular lens and was dubbed crystallin because of its role in maintenance of lens transparency.2 It is also essential for maintenance of micr...
متن کاملThe Zeb proteins δEF1 and Sip1 may have distinct functions in lens cells following cataract surgery.
PURPOSE Posterior capsular opacification (PCO), the most prevalent side effect of cataract surgery, occurs when residual lens epithelial cells (LECs) undergo fiber cell differentiation or epithelial-to-mesenchymal transition (EMT). Here, we used a murine cataract surgery model to investigate the role of the Zeb proteins, Smad interacting protein 1 (Sip1) and δ-crystallin enhancer-binding factor...
متن کاملPrenatal lens development in connexin43 and connexin50 double knockout mice.
PURPOSE To determine the roles of intercellular communication in embryonic eye growth and development, mice with a targeted deletion of the Cx43 gene were examined, and mice without both Cx43 and Cx50 were generated and analyzed. METHODS Embryonic eyes and lenses from wild-type mice, or mice deficient in Cx43, Cx50, or both Cx43 and Cx50 were collected and analyzed structurally by light and e...
